Zbtb1 gene promotes lymphoid development and inhibits myeloid generation in mice

Abstract:Transcription factor Zbtb1 is required for normal lymphoid development.In the present study,we investigate the mechanisms by which Zbtbl promotes lymphoid development and the impact of Zbtb1 on myelogenesis.Specific genes were over-expressed in hematopoietic stem cells by retroviral transduction,and then the hematopoietic stem cells were differentiated in vitro by OP9/OP9-DL1 system.The overexpression of anti-apoptotic gene Bcl2 in the hematopoietic stem cells of Zbtb1-mutant (ScanT) mice only partially recovered early T-cell development,without reverting myeloid bias.In addition,overexpression of the Zbtb1 gene in the wild type hematopoietic stem cells could suppress myeloid development under myeloid-inductive conditions.Taken together,Zbtb1 gene promotes lymphoid development possibly by suppressing apoptosis,while inhibits myeloid development by modulating the transcriptional level of Csf1r.
