Effects of Mycobacterium tuberculosis Rv2986c on dendritic cells maturation and function

Abstract:This study was designed to investigate whether Rv2986c,a newly identified antigen of Mycobacterium tuberculosis,can mature mouse dendritic cells and induce polarization of na1ve CD4 + T cells.The Rv2986c was expressed and purified,and used to stimulate dendritic cells.Then flow cytometry was used to detect the expression level of MHC-Ⅱ surface molecules on the dendritic cells,while ELISA was used to test the secretion levels of IL-6 and IL-12p40 in culture supernatant.In addition,the stimulated dendritic cells was co-cultured with na(i)ve CD4+ T cells,and the secretion level of IFN-γ in the supernatant was tested by ELISA.Data showed that the expression level of MHC-Ⅱ increased significantly on Rv2986c-stimulated dendritic cells.Moreover,ELISA showed that the secretion of IL-6,IL-12 p40 and IFN-γ enhanced obviously after Rv2986c stimulation.In conclusion,Rv2986c can activate mouse dendritic cells to present antigen and contributes to Th1 type polarization of na(i)ve CD4+T cell.
