Effects of long non-coding RNA MALAT1 on macrophage polarization

Abstract:This study was performed to evaluate the effect of long non-coding RNA(lnc RNA)metastasis-associated lung adenocarcinoma transcript 1 (MALAT1) on macrophage polarization.THP-1 cells were induce to differentiate into macrophages by PMA,and then induced to polarize to M1 by IFN-γand LPS,or to M2 by IL-4.The expression of lncRNA MALAT1 in M1 and M2 macrophages were analyzed by real-time quantitative PCR (RT-qPCR).MALAT1-siRNA oligo and NC oligo were transfected into THP-1 macrophages,and the THP-1 macrophages were induced to polarize to M2 by IL-4.Then,the mRNA expression levels of CXCL10,CXCL11,HLA-DR,CCL17,CCL18 and CD163 were detected by RT-qPCR;TNF-α,IL-12,CCL22 and IL-10 secretion induced by IL-4 were detected by enzyme linked immunosorbent assay (ELISA).Data showed that the expression level of MALAT1 in M2 macrophages were significantly higher than that in THP-1 macrophages (MΦ) and M1 macrophages;MALAT1 expression was increased when M1 converted to M2,whereas decreased when M2 converted to M1.Knockdown of MALAT1 following the activation of THP-1 macrophages using IL-4 diminished the expression of M2 markers,and elevated the expression of M1 markers.Compared to the control group,MALAT1 inhibited the increase of CCL22 and IL-10 secretion,but promoted the increase of TNF-α and IL-12 secretion in M2 macrophages.All the result indicate that LncRNA MALAT1 plays an important role in regulating macrophage polarization,knockdown of MALAT1 expression inhibits M2 macrophage polarization,and promote M 1 macrophage polarization.
