The change of cytokines and negative costimulatory molecules PD-1 and Tim-3 in patients infected with Brucella

Abstract:This study was performed to investigate the changes of PD-1 and Tim-3,as well as cytokines IL-17A and IL-6 in patients infected with Brucella.The expression of PD-1 and Tim-3 in peripheral blood mononuclear cells were detected by flow cytometry in 72 patients infected with Brucella and 50 healthy controls;the levels of PD-1 mRNA and Tim-3 mRNA in peripheral blood mononuclear cells were detected by qRT-PCR,and the levels of IL-17A and IL-6 in serum were measured by CBA;the correlations of PD-1 +Tim-3+CD4+T with IL-17A and IL-6 levels in patients infected with Brucella were analyzed by Pearson correlation test;the expression of PD-1 +Tim-3+CD4+T in peripheral blood and the clinical characteristics of patients with Brucella were analyzed.Compared with healthy controls,PD-1 +Tim-3 +CD4+T,PD-1 mRNA and Tim-3 mRNA expression,IL-17A and IL-6 levels were significantly increased (P<0.01) in patients infected with Brucella;pearson analysis showed that PD-1 + Tim-3 + CD4 +T is positively correlated with both IL-17A (r=0.71,P<0.01) and IL-6 (r=0.83,P<0.01).And the expression of PD-1 + Tim-3 + CD4 +T in the peripheral blood of Brucella patients with fever,arthrodynia and multiple organs damages increased significantly (P<0.01).In conclusion,the increased expression of PD-1 and Tim-3 in brucellosis patients may be involved in the negative regulation of Brucella on immune response,which is not conducive to the effective removal of Brucella.
