Neuroprotection of sodium houttuyfonate in spinal cord injury rat and its mechanism

Abstract:The study was performed to investigate the neuroprotection effect of sodium houttuyfonate (SH),the polarization of microglia/macrophages and the microglia/macrophages meditated neuroinflammation in the early spinal cord injury (SCI) in rats.The model of SCI on rats was adopted,and the optimum dose of SH was confirmed by BBB scores,Nissl staining and the survival of motor neurons.The possible neuroprotection mechanisms of SH was evaluated by exploring the expression of TNF-α,IL-1β,P-ERK,ERK P-JNK,JNK,P-P38MAPK,P38MAPK and the M1 and M2 phenotypes microglia/macrophages.Data showed that SH had a certain neuroprotection in SCI rats and the optimum therapeutic dose of SH in this study was medium dose SH 0.12 g· kg-1 · d-1.Compared with SCI group,medium dose of SH treatment could significantly reduce the expression of TNF-α,IL-1β,P-JNK/JNK,P-ERK/ERK,P-P38MAPK/P38MAPK and iNOS,while upregulate the expression of Argi1.In conclusion,SH has a certain neuroprotective effect on early SCI in rats,which may be achieved by inhibiting the activation of MAPK signaling pathway,and then promoting the inflammatory M1 microglia/macrophages transformed to anti-inflammatory M2 cells,thus inhibiting the microglia/ macrophages meditated neuroinflammatory after SCI.
