Immunological characteristics of uterine tissue-resident memory T cells

Abstract:This study was performed to explore the immunological characteristics including distribution,phenotype and function of tissue-resident memory T cells (Trm) in murine uterus by flow cytometry.Balb/c mice were perfused in heart to remove the circulating cells including T cells,then the lymphocytes were obtained by digestion with collagenase and DNase Ⅰ.The spleen tissue was harvested to obtain single cell suspension.Flow cytometry was used to detect the expression of IFN-γ in uterus and spleen when stimulated with PMA + Ionomycin for 5 hours.And the expression of IFN-γ in uterus was compared with that of spleen tissue.Data showed that T lymphocytes in the uterus and spleen of mice express the Trm surface markers CD69 and CD103,and the expression of CD69 and CD103 in the uterus was much higher than that of the spleen cells.Furthermore,CD69+ T cells expressed higher IFN-γ than CD69-T cells.Whereas CD103-T cells expressed higher IFN-γthan CD103+T cells.The expression of memory cell surface molecule CD44 in Trm ceils (CD69+CD103+ and CD69+CD103-T cells) was much higher than that in CD69-CD103-and CD69-CD103+T cells.In conclusion,there is a group of Trm cells in the uterus of mice,which express CD69 and/or CD103 on the surface,and CD69+T cells express higher IFN-γ than CD69-T cells,and CD103-T cells express higher IFN-γthan CD103+T cells.
