The functional changes of γδ T cells in umbilical cord blood of premature infants

Abstract:To explore the significance of γδ T cell function changes in resisting the virus infection in preterm infants,we analyzed the changes in the number,proportion and related function of γδ T cells in umbilical cord blood of preterm infants and term infants.The percentage of γδ T cells in umbilical cord blood mononuclear cells (CBMC) was detected by flow cytometry.The CBMCs from preterm and term infants were stimulated by infection of the influenza virus for 24 h,and then the cytokine production of Perforin,IFN-γand TNF-α by γδ T cells were tested by flow cytometry.The comparison of amplification efficiency of γδ T cells in CBMCs was performed between premature and term infants,followed by stimulation of IPP and IL-2.Data showed that the proportion of γδ T cells in CBMCs of preterm infants was significantly less than that of term infants;the expression of Perforin,IFN-γand TNF-α by γδ T cells increased obviously both in preterm and term infants,but the extent was bigger in term infants.After stimulation by IPP and IL-2,the proliferation of γδ T cells in CBMCs increased both in preterm and term infants,however which was significantly lower in preterm infants than that of term infants.In conclusion,the deficiencies existing in the number and function of γδ T cells in preterm infants may be one of the important factors of being easier to suffered from infectious diseases in premature infants.
