Regulatory effects of purinergic p2x7 receptor on ISG20 in THP-1 macrophages

Abstract:The aim of this study is to investigate the impact of the purinergic p2x7 receptor (p2x7R) on expression of interferon-stimulated gene products of 20 kDa (ISG20) in THP-1 macrophages induced by lipopolysaccharide (LPS).THP-1 macrophages exposed to LPS for specific times were separated into different groups treated with nonspecific antagonists,specific antagonists,or specific agonists,and cultured with IFN-α for 24 h.The caspase-1 inhibitor was added in the groups 1 h before BzATP and IFN-α.mRNA expression of ISG20 in cells treated with the different reagents was determined by RT-PCR and Western blotting;the interleukin-1β in supernatants was detected by ELISA.Data showed that the ISG20 mRNA induced by IFN-α was reduced significantly in the groups treated with the nonspecific antagonist and the specific antagonists (P<0.05),and elevated by BzATP (P<0.05);ISG20 protein expression was upregulated gradually with the increase of activator,but downregulated in BBG.On the other hand,we observed that the IL-1β concentration in the supernatants was correlated with the concentration of BzATP and BBG.Meanwhile,caspase-1 inhibitor (AC-YVAD-cmk) could interrupt the expression of the ISG20 protein induced by BzATP,but not in the IFN-α groups.These results demonstrated that ISG20 mRNA and protein expression could be modulated by the status of the p2x7R and attenuated by the caspase-1 inhibitor via a pathway different from the IFN-α.This finding could aid identification of new therapeutic targets for viral infection.
