The function of Lnc-CLRN3 and its correlation with intestinal innate immunity

Abstract:In order to study the functions of a new long-noncoding RNAs (lncRNAs) in innate immunity,Human colorectal cancer cell line SW480 cells were exposed to LPS stimulation for 4 hours,and then the cells were harvested and subjected to RT-qPCR for screening the LPS-sensitive lncRNAs,and Lnc-CLRN3 was selected.The function of Lnc-CLRN3 was analyzed by gene over expression and knockout,meanwhile,Dual luciferase assay,RNA co-immunoprecipitation and other technical methods were used to analyze the gene function.Data showed that Lnc-CLRN3,highly expressed in SW480 cells and up-regulated by LPS stimulation,was mainly distributed in nucleus,and its transcription was regulated by MAPK pathway,and this gene was combined with SFPQ to control the downstream IL12P40 gene expression.In conclusion,Lnc-CLRN3 is involved in the innate immunity regulation of intestinal cells.
