Preparation and identification of monoclonal antibody against GP2 of lymphocytic choriomeningitis virus

Abstract:The purpose of this study is to prepare monoclonal antibody (mAb) against lymphocytic choriomeningitis virus (LCMV).We utilized the technical of cell hybridization to obtain hybridoma,and the medium of hybridoma was collected.Then the subclass of mAb CH and the immune potency were identified via immunofluorescence (IFA),dot-blot and ELISA.Subsequently,the mAb was purified by protein G Sepharose affinity chromatography protocol,and the purity of the mAb was checked by SDS-PAGE.At last,Western-blot was performed to detect the site where the mAb interacted with the LCMV antigen.As a result,we obtained a hybridoma strain which could secreted specific anti-LCMV mAb stably,and a kind of purified mAb named Anti-GP2 was collected favorably.IFA and ELISA demonstrated that the mAb could recognize the antigen from LCMV.ELISA result indicated that the subclass of the mAb CH is IgG2b,and the immune potency of the medium is 1.35 × 103 approximately;SDS-PAGE showed that the mAb was purified by protein G Sepharose affinity chromatography protocol successfully;Western-blot implied that the site where the mAb combined specifically was GP2 on surface of the LCMV particle.In conclusion,we has prepared a kind of mAb successfully,which could recognize GP2 on surface of the LCMV particle specifically.
