Analysis of immunological repertoire in the peripheral blood of chronic hepatitis B patients with clearance of HBsAg after IFN-α treatment

Abstract：To find the characteristics of the immunological repertoire in the peripheral blood of chronic hepatitis B patients with clearance of HBsAg after IFN-α treatment,we collected peripheral blood of chronic hepatitis B patients to get the lymphocytes by density gradient centrifugation.After IFN-α treatment,the patients with or without clearance of HBsAg were selected,and their corresponding lymphocytes were picked up for total RNA extraction.Then the total RNA was reverse transcribed to cDNA,and the CDR3 sequence of TCRβ was amplified,which was compared with IMGT database then analyzed.Data showed that the diversity of CDR3 of the peripheral blood T cell was lower in chronic hepatitis B patients with clearance of HBsAg as compared with chronic hepatitis B patients without clearance of HBsAg.Furthermore,there were also difference in other immunological features,such as the usage of V/J gene,the pair of V and J gene,the insertion of gene and CDR3 length,except the deletion of gene.So,we can draw a conclusion that chronic hepatitis B patients with clearance of HBsAg after IFN-α treatment display distinct immunological repertoires,which could be used in clinical outcome prediction of chronic hepatitis B patients.
