The role of Th17 cell in renal ischemia-reperfusion injury

Abstract:This study was conducted to explore the role of Th17 cell in renal ischemia-reperfusion injury (RIRI).We established mouse models of RIRI and collected blood,kidney and spleen samples from the mice at indicated time points after RIRI.Renal function was monitored by measuring serum creatinine at different time intervals after reperfusion,and the renal pathological changes were examined by PAS staining.The number of infiltrating lymphocytes was counted.CD4+ T-cell subsets in the kidney and spleen were analyzed by flow cytometry.Data showed that 6 h after reperfusion,there was a slight damage in the kidney,while the number of infiltrated lymphocytes was the most.Surprisingly,24 h later,the number of lymphocytes decreased to normal level whereas RIRI mice exhibited severe kidney structure damage.We further observed that CD4+ T lymphocytes were increased in the kidney and spleen after RIRI,especially the subgroup of Th17 cells.In contrast,there was no change in the percentage of Th1 and Th2 cells after RIRI.These findings suggest that the infiltrating of lymphocytes present a "lag effect" in the early phase of RIRI.Th17 cells,but not Th1 and Th2 cells may play an important pathophysiologic role in renal ischemia-reperfusion injury.
