Autocrine production of IL-6 promotes adhesion and invasion of ovarian cancer cells by Ras/MEK/ERK and PI3K/Akt pathways

Abstract:This study conducted to discover the effect of endogenous interleukin-6 (IL-6) in ovarian cancer cells on cell adhesion and invasion as well as the related signal pathways.Based on our previous work,overexpression of IL-6 in non-IL-6-expressing A2780 cells (by transfecting with plasmid encoding sense IL-6) enhanced cell adhesion and invasion,while depletion of endogenous IL-6 expression in IL-6-overexpressing SKOV-3 cells (by transfecting with plasmid encoding antisense IL-6) reduced the above effects.Furthermore,overexpressing or deleting of IL-6 in ovarian cancer cells increases or decreases basal phosphorylation levels of both ERK and Akt,and additional treatment with specific inhibitor of the ERK or Akt signalling pathway significantly inhibited the adhesion and invasion of IL-6-overexpressing A2780 cells.All above data showed that IL-6-increased ovarian cancer cell adhesion and invasion correlated with activation of Ras/MEK/ERK and PI3K/ Akt.These results suggest that IL-6 secreted by ovarian cancer cells can promote adhesion and invasion.Therefore,modulation of IL-6 expression or its related signaling pathways may be a promising strategy for controlling the progression of ovarian cancer.
