Inhibitory effects of amniotic fluid-derived mesenchymal cells on the proliferation of peripheral blood mononuclear cells and its mechanism
Abstract:In this study, we would identify the amniotic fluid-derived mesenchymal cells (AF-MCs) and explore the mechanism of the AF-MCs suppressing the proliferation of peripheral blood mononuclear cells (PBMC). We obtained and cultured the AF-MCs through the selective digestion and clone cultivation. Then FACS and Giemsa staining were employed to analyze the karyotype and phenotype of the AF-MCs for identifying the cells source and characters. And this analysis showed that AF-MCs were derived from the exfoliated cells during the embryonic development, and had the phenotype familiar with the BM-MSCs. We co-cultured the AF-MCs with PBMC, and FACS analysis showed that the AF-MCs have the immuno-suppression function by secreting the TGF-beta to induce the CD4~+CD25~- T cells into the CD4~+CD25~+Foxp3~+ T cells. Above results imply that AF-MCs are familiar with the BM-MSCs in the phenotype and immunomodulation. So they are a new and secure seed cells for the clinical application of MSCs.
