Establishment and comparison of ELISA, TRFIA, and ALPHAScreen as assay methods for mouse TNF-α

Abstract：To establish and evaluate enzyme-linked immunoassay (ELISA), time-resolved fluoroimmunoassay (TRFIA) and AL-PHAScreen for the detection of mouse TNF-α. Two specific monoclonal antibodies were selected to constructed mTNF- α -ELISA, mT-NF-α-TRFIA, and mTNF-α-ALPHAScreen; the assay conditions of three methods were optimized and they ,sere compared with respect to kinetics, assay parameters, and correlation. The data showed ALPHAScreen was the least time consuming, while TRFIA and AL-PHAScreen had the best sensitivity and dynamic range. The three methods also showed highly significant correlation. The mTNF- α -TR-FIA and mTNF-α-ALPHAScreen methods developed in this study are sensitive, stable, and could meet the need of application.
