Relation between phosphate and immunopotentiation of recombinant HEV-aluminum hydroxide adjuvant vaccines

Abstract:To explore the relationship between phosphate (PO)_4~(3-) and immunogenicity of HEV-aluminum hydroxide adjuvant vac-cine, and to investigate the association of antibody titers with adsorption percentage of antigen in sheep lymph fluid. We prepared ex-perimental vaccines with aluminum-hydroxide adjuvant, which has been pretreated with different quantity of phosphate. The experi-mental vaccine was used to immunize Balb/c mice intraperitoneally (i.p.). Subsequently, the immune sera at different weeks were col-lected and the antibody titer was measured quantitatively by ELISA. Phosphorus in precipitate was determined by ammonium molyb-ed according to the langmuir adsorption equation. And adsorption percentage of antigen was quantitatively detected by ELISA when percentage of antigen decreased after the mix of vaccine with sheep lymph fluid, antibody titer of immune mouse sera raised gradually until steady state. The results suggest that increasing of phosphate content in certain extent of vaccine can decrease adsorption per-centage of antigen, reduce antigen adsorption on aluminum adjuvant, but enhance antibody titer.
