Localization of conjugate vaccines PPS14-OVA and PPS14-C3d in mouse spleen and effects of complement on localization of the conjugate vaccines

Abstract:To investigate the localization of conjugate vaccines consisting of serotype 14 pneumococcal capsular polysaccharide (PPS 14) coupled to either ovalbumin (OVA) or C3d in mouse spleen cells, and to evaluate the role of complement in localization of the con-jugate vaccines. Mice were immunized with PPS14-OVA or PPS14-C3d by either subcutaneous or intravenous injection, while some mice pretreated with cobra venom factor (CVF). Immunofluoresecnce was used to examine the frozen sections of spleen from all mice for detecting PPS14 and to identifying various immune cells. Immunofluorescence illustrated that PPS14-OVA was bound to B cells in the spleen marginal zone within 1 hour, first appeared in the follicle at 2 hours, and localized solely in the follicle at 6 hours after immunization; the antigen was conjuncted with follicular B cells and follicular dendritic cells, but not with macrophage or T cells. CVF treatment completely abolished the localization in the spleen of PPS14-OVA, but not of PPS14-C3d, regardless of immunization routes. All these data together suggest that complement is required for PPS14-OVA localization, but not PPS14-C3d, to the spleen.
