Humoral immune response against HER-2 induced by immunization with peptide fusion protein

Abstract:To induce the humoral immune response against peptide Mut (a mimic B-cell epitope of HER-2) and HER-2 in mice immunized with the fusion protein of heat shock cognate protein 70 (Hsc70) and peptide Mut. The DNA encoding Hsc70 and peptide Mut was inserted into pQE40 vector to construct prokaryotic expression vector pQE40-HscTO-Mut, and then expressed in E.coli SG13009. The Hsc70-Mut fusion protein was purified with Ni-NTA resin and the purity re.hod more than 90%. After Balb/c mice were immunized with the fusion protein, the anti-peptide Mut and anti-HER-2 humoral immune response were measured by ELISA and Western blotting, respectively, and found that 1 in 6 mice could generate antibody to HER-2. As a result, we conclude that peptide Mut has potential for being developed as a HER-2 vaccine for biotherapy of cancer with HER-2 overexpression. Moreover, immunizing with peptide fusion protein is a strategy for peptide immunization.
