Effects of T-reg cell on cytotoxicity of NK cells against breast cancer cell in vitro

Abstract:To explore the effects of T-reg on NK eytotoxicity against breast cancer cells and the possible mechanism.Flow cytometry (FCM) was used to evaluate the proportion of T-reg, NK cell, and T cell subsets in peripheral blood of breast cancer patients; lactate dehy-drogenase (LDH) assay was manipulated to test the cytotoxicity of NK cell on breast cancer cells; EL1SA was performed to examine the level of IFN-γ secreted by N K cell and TGF- β1 secreted by T-reg cell in supernatant.The proportion of T-reg cells in patients with breast cancer was higher than that of volunteers (7.5% ± 3.0% vs 5.1% ± 1.5%, P < 0.01).T-reg cells inhibited the cytotoxicity of NK cell on breast cancer cell and affected the secretion of IFN-γ by NK cells, while TGF-β1 level in the supernatant increased with the elevation of T-reg proportion.The results show that T-reg cells may inhibit the cytotoxicity of NK cell, which might associate with the secretion of TGF-β1
