Mechanism of the effect of insulin on D5 dopamine receptor expression in renal proximal tubule

Abstract:Objective To investigate the mechanism of the effects of insulin on D_5 dopamine receptor expression in renal proximal tubule (RPT).Immortalized RPT cells from wkyrats were prepared for this experiment.Immunohistochemical method was used to detect the effect of insulin on D_5 receptor expression, while immununoblottings were performed to confirm the impacts of genistein, wortmannin and PKC inhibitor peptides 19-31 on the effect of insulin on D_5 dopamine receptor expression.Insulin increased D_5 receptor protein expression in WKY RPT cells.But genistein, wortmannin and PKC inhibitor peptides 19-31 respectively blocked the effects of insulin on D_5 dopamine receptor expression.Insulin can regulate D_5 dopamine receptor expression and the effect might be related with tyrosine protein kinase, PI3K, and PKC.The experiment results suggest that the disbalance between receptors may cause dysfunction of receptors, thus produce the abnormity in natriuresis and immunization in the kidney.
