Human extracellular domain of 4-1BBL enhances the PBL-based antitumor effect of anti-CD3/anti-Pgp diabody in vitro and vivo

Abstract:To study effects of the human extracellular domain of 4-1BBL (ex4-lBBL) on antitumor activity of anti-CD3/anti-Pgp diabody. Ex4-1BBL and anti-CD3/anti-Pgp diabody were expressed in E. Coli strain, and then the expression products were purified with affinity chromatography. Nonradioactived Cytotox 96 assay kit was used to observe the cytotoxicity of PBL regulated by ex4-1BBL and an-ti-CD3/anti-Pgp diabody in vitro with the their target cells (K362/A02) at different E:T ratio. The results demonstrated that ex4-lBBL en-hanced the antitumor immunity of PBL against multidrug resistant K562/A02 transplanted in nude mice. Tumor growth was dramatically in-hibited by the combined therapy of ex4- 1BBL and anti-CD3/anti-Pgp. Our results demonstrated that soluble ex4-1BBL, as a immunoadju-vant, can modulate the PBL activation and enhance the outcomes of PBL-based antitumor biotherapy. Thus, ex4-lBBL may become a new agent for bio-immunotherapy.
