Immunogenicity of combined B-cell epitope of tumor antigen MAGE-12 immunogen

Abstract：Objective To design and synthesis B-cell epitope of tumor antigen MAGE-12 and investigate its immunogenicity.Methods Based on epitope MAGE-12_(82-93) and MAGE-12_(3-14),a multiple antigenic peptide system(MAP)was synthesized as immunogen.Three groups C-57BL/6 mice were immunized with the immunogen of MAGE-12_(82-93),MAGE-12_(3-14),and MAGE-12_(3-14)+MAGE-12_(82-93),respectively.Thus the polyclonal anti-serum was obtained,and then identified by using an ELISA and immunohistochemical staining.Results The antibody titers of the three groups were reached 1:64,1:256,1:512,respectively,while the titer of polyclonal antibody from MAGE-12_(3-14)+MAGE-12_(82-93) group was the highest.The polyclonal antibody was identified as specific antibody of the antigenic peptide.Conclusion The immunogen based on MAGE-12_(3-14)+MAGE-12_(82-93) is stronger than that of other single B cell epitope of MAGE-12.
