The expression of PINCH-1 in leukemia bone marrow stromal cells and its significance

Abstract：Objective To investigate the expression of PINCH-1 in acute leukemia bone marrow stromal cells and its significance.Methods Bone marrow stromal cell were cultivated.The expression of PINCH-1 mRNA and protein were observed by RT-PCR and Western blotting;the distribution and expression of PINCH-1 protein in target cells were observed by immunocytochemistry and immunofluorescence technique.Results The levels of PINCH-1 mRNA and protein were significant higher in bone marrow stromal cells of patients with acute lymphoblastic leukemia(ALL)and acute myeloid leukemia(AML),as compared with those of normal bone marrow stromal cells(P＜0.01,P＜0.01).The levels of PINCH-1 protein have significant disparity between AML group and ALL group (P＜0.01),but the levels of PINCH-1 mRNA similar between them.Conclusion The expressions of PINCH-1 in leukemia bone marrow stromal cells are significantly higher than that in normal bone marrow stromal cells,implying high level of PINCH-1 in leukemia bone marrow stromal cells may related with the generation and progression of acute leukemia.
