Silencing breast cancer cell Livin gene in vitro

Abstract:Objective To study the suppressing effects of Livin-siRNA on the Livin gene expression in MCF-7,ZR-75-30 cells,as well as on the proliferation of breast cancer cells.Methods Three siRNA eukaryotic expression vectors targeting Livin were constructed and transfected into breast cancer cells.The Livin mRNA expression and the Livin protein expression in breast cancer cells were examined by real-Time quantitative PCR and western blotting,respectively.The suppressing effects on proliferation of MCF-7,ZR-75-30 cells were detected by MTT assay.Results Three specific siRNA eukaryotic expression vectors targeting Livin were successfully constructed,which could inhibit the expression of Livin mRNA and protein to varying degrees,and si-Livin2 resulted in the highest inhibiting rate.The Livin mRNA expression and its protein expression were decreased significantly in MCF-7 and ZR-75-30 breast cancer cells after si-Livin2 treatment(P<0.05).Forty-eight hours after si-Livin2 transfection,the proliferation of cells was strongly inhibited.Conclusion Silencing Livin could inhibit the MCF-7 and ZR-75-30 cells Livin mRNA and its protein expression and suppress proliferation of breast cancer cells.
