Effects of dermatophagoides farinae(Der.f)extract on DC2.4

Abstract：Objective To investigate the effects of Dermatophagoides farinae(Der.f)extract on DC2.4.Methods Interleukin-12(IL-12)and IL-10 production,TLR2 and TLR4 gene expression,as well as activation of p44/42 mitogen-activated protein kinase(MAPK)of DC2.4 were assayed,respectively.Results Der.f extract induced DC2.4 to enhance IL-10 secretion(from 0 in control group to 27.7 pg/mL in Der.f treatment group,P＜0.01),but reduce IL-12 secretion(from 22.8 pg/mL in control group to 15.9 pg/mL in Der.f treatment group,P＜0.05).Der.f extract also enhanced phosphorylation of p44/42 MAPK.But U0126 could reverse the Der.f extract-induced cytokines production patterns and suppressed the phosphorylation of p44/42 MAPK.Conclusion Der.f extract may affects cytokines secretion,gene expression,and signal pathway of DC2.4.
