Immune regulatory effects of genistein on animal model of rheumatoid arthritis

Abstract:Objective To explore immune effects of genistein on rheumatoid arthritis(RA).Methods The Ⅱ collagen-in-duced arthritis(CIA)model was established with mice;mice were devided normal group,model group,high-dose group genistein,mild-dose genistein group,low-dose genistein group,and glucosidorum tripterygll totorum group.The CIA mice were invaluated for observing T lymphocyte proliferation,T lymphocyte adhesion ability,primary synovial cell proliferation,and effects of cell culture supernatants on MMP2 and TIMP-2 expressions in spleen cells.Results In mid/high-dose genistein group,under the conditions of RPMI-1640 medium and ConA stimulating,all detecting indexes were significantly decreased,including spleen cells stimulation index,level of spleen cell adhesion to collagen type Ⅱ,sensitized T-lymphocyte adhesion ability/percentage,the primary synovial cell proliferation ability,and levels of IL-1β and TNF-α in synovial cell culture supematant.Genistein allso could step-up the expression of MMP2 and cut down the expression of TIMP-2 on the spleen cells of the CIA mice.Conclusion Genistein plays immune regulatory role in RA treatment,with better clinical effects but lower side-effects.
