Comparison of CD8~+ NKT cells and CTL in anti-tumor effects

Abstract：Objective To establish a new method for inducing CD8~+ NKT cells in vitro and evaluate the antitumor ability in vitro.Methods Tumor cells(EG7-OVA or B16-F10)were stained with fluorescent dyes,and then co-cultured separately with CD8~+ NKT cells and CTL,which were sorted by flow cytometry depending on their surface markers and had been activated with peptide-load-ed-DCs in vitro for 2 weeks.After 12 hours,target cells were stained with PI and Annexin V.and were analyzed by flow eytometry.Resuits CD8~+ NKT cells could kill EG7-OVA and B16-F10 in a direct cytotoxic manner.Conclusion CD8~+ NKT cells have both antigen specific and non-specific effects on tumor cells.Due to such a dramatic anti-tumor effects of CD8~+ NKT cells.its interruption should be excluded when studying the function of CTL.
