Purification and characterization of glyceraldehyde-3-phosphate dehydrogenase from Lactoba.cillus acidophilus

Abstract:Objective To purify and characterize glyceraldehyde-3-phosphate dehydrogenase (GADPH) from L. acidophilus ATCC 4 356. Methods GADPH was purified through procedures of ammonium sulphate fractionation, affinity chromatography, anion-exchange, and sieve chromatography. Then the purified product was characterized by N-terminal sequencing. The apparent molecular weight of homogenous GADPH was measured by molecular sieve. Results The purified product showed a single band of 3.7×104 and was identified as GADPH of L acidophilus by N-terminal sequencing. The apparent molecular weight measured using molecular sieve was 7.08×104, which indicated the GADPH was dimeric. Conclusion The homogenous GADPH of L. acidophilus could be abundant-ly obtained by the purified program, which lays a foundation for the study of its immune function and using as an immune enhancer in the future.
