Haematopoietic negative regulatory factors in patients with paroxysmal nocturnal hemoglobinuria and aplastic anemia

Abstract:Objective To detect the expression levels of IFN-γ, TNF-α, and TGF-β1 in peripheral blood plasma of patients with paroxysmal nocturnal hemoglobinuria (PNH) and aplastic anemia (AA) for studying the relationship of the expression levels of IFN-γ, TNF-α, TGF-β1 with the pathogenesis of PNH and AA. Methods Peripheral blood was collected from 18 patients with PNH,20 patients with AA and 15 controls. Enzyme-linked immunosorbent assay (ELISA) was used to measure the expression levels of IFN-γ, TNF-α, and TGF-β1in plasma. Results The IFN-γ and TNF-α levels in AA patients were higher than those in controls (P <0.05 ) ; the TGF-β1level in AA patients was much higher than that in controls (P<0.01). The IFN-γ, TNF-α, and TGF-β1 levels in PNH patients were much higher than those in controls (P<0.01). The TNF-ot level in PNH group was higher than that in AA group (P <0.05 ), while the IFN-γand TGF-β1 levels in PNH group were much higher than those in AA group (P<0.01). Conclusion The results suggest that the haematopoietic negative regulatory factors, such as IFN-γ, TNF-α and TGF-β1 , could inhibit hematopoietic cells in PNH and AA patients, which imply that the parasecretion of cytokines may contribute to the pathogenesis of PNH and AA.
