Ovarian reserve function in reproductive aging after Pregnant Mare serum Gonaaotropin stimulation

Abstract:Objective To measure serum anti-Mullerian hormone (AMH) levels in young, adult, and aging rats after PMSG stimulation. Methods Female SD rats were divided into young, adult, and aging groups according to their age. After PMSG stimula-tion, serum AMH levels were measured by ELISA. AMH positive follicles were showed by immunofluorescent staining. Results Serum AMH levels were (2.92±0.55 ) ng/mL, ( 1.90±0.11 ) ng/mL, and (0.80±0.08) ng/mL in young, adult, and aging rats, respec-tively, which was significant differences (P<0.05, young vs adult; P <0.001, young vs aging; P < 0.001, adult vs aging) ; total folli-cle counts were 57.3±5.5, 31.6±3.6, and 15.1±1.4 in young, adult, and aging rats, respectively, which was significant differ-ences (P<0.001, young vs adult; P<0. 001, young vs aging; P<0.05, adult vs aging) ; the numbers of AMH positive follicles were 9.2±2.0, 6.3±1.4, and 4.4±0.8 in young, adult and aging rats, respectively. Conclusion After PMSG stimulation, AMH-se-creting follicles and serum AMH levels are decreased significantly according to aging. AMH could be a good biological marker to predict ovarian reserve even after PMSG stimulation.
