Vasoactive intestinal peptide modulates the immune function of LPS-stimulated dendritic cells

Abstract:Objective To investigate the modulating effects of vasoactive intestinal peptide (VIP) on the immune function of LPS-stimulated dendritic cell. Methods DCs were prepared from bone marrow of C57BL/10.A with rmGM-CSF and rmIL-4, and then stimulated with different stimuli for different times. The stimulated DCs and supematant were collected for ELISA analysis; total RNA was extracted from the stimulated DC and analyzed by RNAse protection Assay. Results VIP significantly inhibited LPS-stimulated DC to produce IL-2, IL-12, TNF-α (P<0.05), MIP-2, and IP-10 (P<0.05), but the down-regulation of MIP-1α, MIP-1β, and Rantes produced by LPS-stimulated DC was not significant (P>0.05). Conclusions VIP negatively modulates the immunity of LPS-stimulated DC.
