Expression of Toll signal pathway-associated genes in bladder cancer cell line stimulated by polyporus polysaccharide

Abstract:Objective To explore the expression of Toll signal pathway-associated membrane molecules in mouse bladder canc-er cell line T739 stimulated by polyporus polysaccharide(PPS). Methods PPS was added to culture with T739 at final concentration of 0.25 mg/mL; relative quantitative RT-PCR was used to measure the levels of CDI4, TLR2, MD-2, Mydd88, TRAF6, TIRAP, I-RAK1, TOLLIP, NFKB1, NFKB2, Rel A, CHUK, CCL2, ICAMI, ECSIT, MAP3KI, and IKBKB mRNA using SYBR Green fluoro-chrome at variable time points. LPS group and blank group were set as controls. Results PPS was significantly affected the expression levels of CD14, TLR2, MD-2, NFKB1, CHUK, MAV3K1 and IKBKB mRNA, whose ratios were 2.85, 3.12, 4.74, 3.39, 4.45,40.25 and 0.07 respectively; however other mRNA has no significant change after treatment of PPS. Only CD14, TLR4, ICAM1,NFKBI, and IKBKB mRNA were increased after the stimulation of LPS. No significant ahange of all mRNA levels was found in blank group. Conclusion Toll signal pathway-associated genes are activated in urothelial cell by PPS, possibly through TLR2. Decrease of IKBKB expression is probably associated with suppressing bladder cancer cell multiplication.
