Apoptosis of peripheral blood mononuclear cells of patients with hepatitis B and its significance
Abstract:Objective To study the significance of AICD(activation-inducedcell death) in the pathogenesis of chronic hepatitis B and chronic severe hepatitis B. Methods Peripheral blood mononuclear cells(PBMCs)were isolated from 10 healthy blood donors and 20 patients with chronic/chronic severe hepatitis B. PBMCs were cultured with PHA-P for 72 hours in vitro. Then apoptosis of the PBMC was assayed by flow cytometry.Results The percentage of PBMC apoptosis of hepatitis B group was significantly higher than that of normal control group [(25.48±14.07)% vs (11.45±5.27)%,P<0.01]. The percentage of PBMC apoptosis of chronic hepatitis B group[(30.57±13.43)%] was also significantly higher than that of normal control group[(11.45±5.27)%, P<0.01]and higher than that of chronic severe hepatitis B group[(13.59±6.44)%, P<0.01]. The percentage of PBMC apoptosis of hepatitis B with HBeAg(+) group was significantly higher than that of normal control group [(29.50±12.54)% vs (11.45±5.27)%], P<0.01].Conclusion The results show that AICD may be an important mechanism of peripheral immune tolerance in HBV chronic infection.
