Discovery of a new allele and polymorphism at the TNFd locus in Hubei Han population
Abstract：Objective To investigate the distribution of TNFd microsatellitepolymorphism in Chinese Han population in Hubei province．Methods DNA samples were extracted from 164 unrelated healthy individuals’EDTA-blood．TNFd microsatellite alleles were typed using PCR technique，followed by High-Voltage denaturing PAGE，with silver staining．At the same time，the PCR products were cloned and sequenced．Results We detected eight alleles and twenty kinds of genotypes at the TNFd locus．A new allele was found and identified．No deviation from Hardy-Weinberg equilibrium was observed．Statistical analysis showed that the distribution of TNFd allele in Chinese Han population was significantly different from that in European or in American Caucasians（P<0.05）．Conclusion There exists ethnic difference in the distribution of TNFd allele．The sequence information of the new allele can be retrieved from the genebank and its accession number is AF315593．
