An unidentified protein tyrosine kinase regulates IL-6-induced Ras/MAPK/NF-IL-6 pathway activation in myeloma cells
Abstract:Objective To investigate the activation and regulation of twoIL-6 signal transduction pathways-JAK/STAT and Ras/MAPK/NF-IL-6 in a human myeloma cell line-KM-3. Methods Firstly, the activation of the transcription factors(STAT3,NF-IL-6) and protein kinases(JAK1,MAPK) involving in IL-6 signal transduction were detected by electrophoretic mobility shift assay(EMSA) and immunoprecipitation(IP) respectively in KM-3 cells stimulated by IL-6.Then the cells were treated with Genistein, a specific protein tyrosine kinase inhibitor, and the changes on the activation of IL-6 signa-ling pathways were also exhibited by EMSA. Results We only observed the activation of Ras/MAPK/NF-IL-6 pathway in KM-3 cells stimulated by IL-6,but JAK/STAT pathway can not be activated at the same conditions. However, Genistein exerted an obvious inhibition effect on Ras/MAPK/NF-IL-6 pathway activation in KM-3 cells. Conclusions An unknown protein tyrosine kinase other than JAK1 can participate and regulate IL-6-induced activation of Ras/MAPK/NF-IL-6 pathway in KM-3 myeloma cells.
