Effect of immunogenicity of liver cancer cell lines by transduction of adenovirus-mediated human B7-1 gene
Abstract:Objective To investigate the change of immunogenicity of livercancer cells transduced with adenovirus-mediated human 4 kind of genes (p53,GM-CSF,B7-1,IL-2).Methods Using human mixed lymphocyte of peripheral blood and tumor cell coculture,cytotoxicity assay of peripheral blood T lymphocyte,we studied the change of immunogenicity of liver cancer cells transduced with Ad-multigenes.Results The proliferation of human peripheral blood T lymphocytes and T lymphocyte cytotoxicity activity against parent tumor cells were increased by transduction of Ad-multigenes in liver cancer cells HepG2 and BEL-7402.IL-12 has a strong synergetic effect with Ad-multigenes in inducing proliferation and cytotoxicity of T cells.Conclusion The immunogenicity of liver cancer cells was enhanced after transduction of adenovirus-mediated multigenes which containing B7-1 molecule.
