Preparation and identification of McAbs against human augmenter of liver regeneration

Abstract:Objective This study was designed to prepare monoclonal antibodies against human augmenter of liver re-
generation (hALR),and establish a sensitive assay for it. Methods Monoclonal antibodies were prepared by hybridoma tech-
nique. Results Four hybridoma,which stably produced monoclonal antibodies (McAbs) against human augmenter of liver re-
generation (hALR),were established by fusion of SP2/0 myeloma cells and splenocytes from Balb/c mice immunized by re-
combinant hALR. The titers of specific antibody in culture supernatants and ascites fluid were 1×10-2and 1×10-5 respective-
ly by indirect enzyme linked immunosorbent assay (ELISA). The four McAbs belonged to IgG1 and IgG2b subclasses respec-
tively and were shown to be reactive with hALR,but not with other cytokines (G-CSF,GM-CSF and IL-6) by ELISA. The
results of immunoblotting analysis indicated that the four McAbs all reacted with hALR. Conclusion These antibodies were
able to specifically react with hALR,and could be further used in other study.
