Abstract：Objective To screen and characterize human phage antibody to hepatitis C NS4A antigen with phage dis
play technique. Methods The recombinant phages were panned by recombinant NS4A antigen which was coated in a mi-
crotiter plate. After three rounds of biopanning,48 clones were determined specific to NS4A antigen. The binding characteriza-
tion of each clone of ScFv phage to HCV NS4A was demonstrated by ELISA method. Results NS4A specific antibody has a
specific combination character with hepatitis C virus NS4A antigen. The DNA sequence data showed that the ScFv gene in-
cluded 771 bp. Conclusion Human single chain antibody to hepatitis C NS4A antigen has been identified by means of the phage
display technology.
