Influence of retinoic acid on CDK2-overexpressing HL—60 cells
Abstract:Objective To determine the function and expression of CDK2 in the course of proliferation and differentia-tion of HL—60 cells induced by retinoic acid(ATRA).Methods We introduced CDK2cDNA into HL—60 cells and treated them with ATRA.Results ATRA could inhibit the growth and induce differentiation of HL-60 cells overexpressing CDK2,about 80 percent cells were arrested at G0/G1 phase after 3d exposure to ATRA.Comparing with the cells without trapsfection,the effect of ATRA on the transfected HL—60 cells was not so muchas that on the normal HL-60 cells.Moreover,the protein ex-pression of CDK2 did not change dramatically after exposure to ATRA. Conclusion The results above suggest CDK2 is very important in G1→Sphase transition,it's overexpression could promote the proliferation.ATRA could inhibit the prolifera-tion and promote the differentiation,but CDK2 overexpression could weaken the effect of ATRA.
