Application progress of CRISPR/Cas9 gene editing technology in the field of animal husbandry science

Abstract:CRISPR/Cas9 gene editing technology,as a new generation of highly efficient and precise genome editing tools,has shown broad application prospects in the field of animal husbandry science.This article systematically expounds the research progress of CRISPR/Cas9 from three dimensions:its technical principles,core applications,and the problems and challenges it faces.The article briefly describes that the technology guides the Cas9 protein to target and cut DNA through single-stranded guide RNA(sgRNA),combined with non-homologous end join(NHEJ)or homologous directed repair(HDR)mechanisms,to achieve gene knockout,insertion or modification.At present,this technology has been successfully applied in disease-resistant breeding[such as editing the Na⁺/H⁺ exchanger 1(NHE1)gene in chickens to resist avian leukemia virus,and knockout of the hemoglobin scavenger receptor(CD163)gene in pigs to inhibit porcine reproductive and respiratory syndrome],production performance improvement[such as myostatin(MSTN)gene editing to promote muscle growth in pigs and cattle],and stress resistance trait improvement[such as uncoupling protein 1(UCP1)gene insertion to enhance body temperature regulation in piglets].Although CRISPR/Cas9 technology has made significant progress,it still faces technical challenges such as off-target effects,the stability of gene editing,and delivery efficiency.In the future,it is necessary to integrate multi-omics analysis,optimize precise editing strategies,and establish an industrialized safety assessment system to promote its sustainable development in the livestock industry.
