Establishment of a Nano-PCR method for detection of Klebsiella pneumoniae from mink

Abstract:This study aimed to establish a sensitive and efficient detection method for Klebsiella pneumoniae of mink origin to improve diagnostic efficiency.Specific primers were designed based on the khe gene sequence of Klebsiella pneumoniae.By optimizing the reaction conditions and thermal cycling program of polymerase chain reaction(PCR),along with specificity/sensitivity tests and clinical sample validation,a Nano-PCR detection method was successfully developed.Results demonstrated that the established Nano-PCR method specifically detected Klebsiella pneumoniae,yielding negative results for Escherichia coli,Shigella spp.,Pseudomonas aeruginosa,Streptococcus spp.,Staphylococcus spp.,and sterile water.The sensitivity of this method was 10-fold higher than conventional PCR.In clinical testing of 30 samples,Nano-PCR achieved a positive detection rate of 70.0%(21/30)for Klebsiella pneumoniae,outperforming conventional PCR(53.3%,16/30).The study confirms that the developed Nano-PCR method exhibits high sensitivity and specificity,proving effective for detecting Klebsiella pneumoniae in minks.
