Isolation,identification,and biological characterization of probiotics from the intestine of Bama Xiang pigs

Abstract:This study aimed to isolate and identify probiotics from the intestines of healthy weaned Bama Xiang pigs to provide a foundation for developing feed additives.Intestinal contents were collected from weaned piglets at Bama Original Xiang Pig Farming and Animal Husbandry Co.,Ltd.Bacteria were isolated using MRS selective medium and identified through hemolysis tests,Gram staining,biochemical characterization,adhesion assays,and 16S rDNA gene homology analysis.Isolates were evaluated for tolerance to simulated gastrointestinal fluid,bile salts,high temperature,and antioxidant activity.The results showed that the four Gram-positive,non-hemolytic strains with hydrophobicity exceeding 90%were isolated.All strains tested positive for lactose,melezitose,mannitol,sorbitol,raffinose,inulin,and sucrose fermentation.16S rDNA sequencing revealed 99%homology with Enterococcus faecalis.The four Enterococcus faecalis strains exhibited a lag phase(0~4 h),followed by logarithmic growth with rapid pH decline.Strain 3-M2 entered stationary phase at 10 h,while strains 3,37,and 39 reached stationary phase at 14 h,with final pH stabilized at 4.0.The survival rate of strain 37 in artificial gastrointestinal fluid,bile salts and high-temperature environment was extremely higher than that of other strains(P<0.01),and its antioxidant capacity was also extremely higher than that of other strains(P<0.01).The isolates exhibited strong hydrophobicity,antioxidant activity,and tolerance to simulated digestion,indicating potential as feed additives for Bama Xiang pigs.
