Isolation and identification of canine parvovirus Shanghai strain

Abstract:In order to study the prevalence of canine parvovirus(CPV)in Shanghai,the intestinal contents of dogs suspected to infect CPV were collected.The virus strain was inoculated and cultured in F81 cat kidney cells in a synchronous inoculation manner,and the fifth generation of virus could produce cytopathic.A virus strain was isolated from the intestinal contents,named as CPV-2/canine/Shanghai/01/2018.The CPV strain was identified by cytopathic observation,electron microscopy morphology test,IF,cell sensitivity test,HA,partial physicochemical properties test and VP2 sequence.The virus titer of TCID50 was 105.3/0.1 mL.It could produce an HA titer of 28.The length of VP2 gene was 1 755 bp.The nucleotide homology and amino acid homology of CPV-2/canine/Shanghai/01/2018 were 98.3%~99.1%and 97.3%~99.3%,respectively.Based on VP2 gene,the strain belonged to CPV-2c type.The study suggests that there are different gene subtypes of CPV in Shanghai,and the correct vaccine strain should be selected in vaccination.
