PCR-RFLP method for cashmere and wool fibre identification

Abstract:Cashmere fibre is more expensive than wool, often adulterated during marketing. So it is very important to find a precise method to identify and differentiate those tow fibers. In this study, a polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) technique was exploited to differentiate cashmere and wool fibres. The DNA in animal hair shafts has been isolated from scoured cashmere and wool fibres. The mitochondrial Cytochrome b sequences of both species were amplified by PCR using primers designed from known sequences. The polymorphism ob-served of the two species were detected by digestion the amplified products by ensonucleases SspⅠ. The RFLP profile clearly distinguishes the cashmere and wool fibres and this method can be ex-ploited to test the quality of cashmere products.
