Abstract:Objective:To detect the expression and location of phosphatase and tensin homolog ( PTEN) deletion on chromosome 10 and Akt proteins in lesions of psoriasis vulgaris , and to inves-tigate their effects on abnormal proliferation of the keratinocytes .Methods:Skin tissue samples were taken from 30 patients with psoriasis vulgaris and 30 healthy controls .The expression of PTEN and Akt was measured by western blot and immunohistochemistry .T-test was used to com-pare expression of PTEN and Akt between the two groups .P-value less than 0.05 was considered to be statistically significant .Results:Immunohistochemical staining demonstrated that both PTEN and Akt were expressed mainly at the location of cytoplasm in the keratinocytes .PTEN showed light yellow granules in psoriatic keratinocytes while dark yellow granules in normal skin keratino-cytes.Akt showed brown yellow granules in psoriatic keratinocytes while light yellow granules in normal skin keratinocytes .Western blot analysis demonstrated PTEN protein was lowly expressed while Akt was highly expressed in psoriatic lesions compared to that of normal controls with charac -teristic significance ( both P<0.05 ) .Conclusion:The down-regulation of PTEN protein and up-regulation of Akt protein may play a role in the hyperproliferation of keratinocytes in psoriatic le -sions.
