Observation of the Therapeutic Effect of Kallikrein-binding Protein on Animal Model of Psoriasis

Abstract:Objective:To investigate the therapeutic effect of kallikrein-binding protein ( KBP) on animal model of psoriasis .Methods:The animal models of psoriasis was established by topical 5%propranolol emulsion liniment on the ear of guinea pigs .Then the established animal models were intraperitoneally injected with KBP or Phosphate Buffer Solution (PBS) respectively, with the normal guinea pigs as experimental control .The changes of ear skin lesions were observed macro-scopically and microscopically .The microvessel density ( MVD) in skin lesions was evaluated by immunohistochemistry .Results:After modeling , scaly erythematous plaques on the ears were ob-served, and the histopathologic changes such as hyperkaratosis , parakaratosis,acanthosis of epi-dermis and capillary hyperplasia as well as inflammatory cell infiltration in dermis were also pres-ent.The MVD in ear skin lesions of modeling group was significantly higher than that of normal group (Z=-2.32,P <0.05).After KBP injection, the psoriatic lesions were obviously im-proved in macroscopic and microscopic view , as both the skin lesion severity scoring and histologic scoring significantly decreased compared to those of experimental controls ( Z=-2.64,-2.66,P<0.05).Furthermore, the MVD was much lower in KBP group than that in PBS group (Z=-2.61,P<0.01).Conclusion:Judged from it’s significant role on inhibiting angiogenesis and improving the psoriatic severity in macroscopic and microscopic view , KBP may be a potential therapeutic agent for psoriasis .
