Expression of EGFR, PTEN, c-Jun and MMP-1 in Facial Seborrheic Keratosis Lesions

Abstract:Objective:To investigate the protein expression levels of EGFR , PTEN, c-Jun and MMP-1, which play an important role in photoaging , in the facial lesions of seborrheic keratosis and to explore the underlying correlation between photoaging and the pathogenesis of seborrheic keratosis.Methods:Specimen from facial lesions of seborrheic keratosis (n=54) and normal fa-cial skin ( n=20 ) was stained with immuno-histochemistry method for the protein expression of EGFR, PTEN, c-Jun and MMP-1.EVG method was performed to reveal elastic fibers and colla-gen fibers.Results:Compared with normal facial skin , the expression of EGFR, c-Jun and MMP-1 in seborrheic keratosis lesions was higher ( Z =-4.45, -2.33, -2.37, respectively, P <0.05 ) , while the expression of PTEN in seborrheic keratosis was slightly lower ( Z=-1.55, P>0.05).The expression of MMP-1 was positively related to EGFR and c-Jun (r=0.71, 0.69, re-spectively, P<0.05) and was negatively related to PTEN (r=-0.31, P<0.05) by Spearman's rank correlation analysis .There was a positive correlation between age and amount of degenera-tive elastic tissue (P<0.05), but a negative correlation between age and amount of collagen fi-bers in seborrheic keratosis than those in controls .Conclusion:It suggests that there is a correla-tion between skin photoaging and the pathogeny of seborrheic keratosis according to the variation of expression about biological markers including EGFR , PTEN, c-Jun and MMP-1, as well as in-creasing degeneration of elastic fibers in the lesions .The occurrence of facial seborrheic keratosis may be generated by photoaging .
