Clinical practice and considerations of anti-CD38 monoclonal antibodies in relapsed/refractory multiple myeloma

Abstract:Multiple myeloma(MM)is a common malignant tumor of plasma cells,accounting for approximately 10%of hematologic malignancies,and its incidence is steadily increasing.Nearly all cases of MM eventually progress to relapsed/refractory multiple myeloma(RRMM),and with the number of treatment lines increases,therapeutic challenges escalate,imposing a substantial social burden.Anti-CD38 monoclonal antibodies,represented by daratumumab and isatuximab,have become a cornerstone of standard therapy.These agents target myeloma cells through multiple mechanisms and have shown high response rates and survival benefits in numerous clinical studies.Nevertheless,their treatment-related adverse effects require standardized management and monitoring.Looking ahead,anti-CD38 monoclonal antibodies continue to offer broad potential for further exploration in areas including combination therapies,novel target development,and formulation innovation.This comprehensive review summarizes the latest research progress,efficacy,safety,and future direction of anti-CD38 monoclonal antibodies in the treatment of RRMM,aiming to provide clinicians with an up-to-date reference.
