Expression levels of microRNA-3163 and microfibrillar associated protein 2 in gastric cancer tissue and their relationship with clinical pathological parameters and prognosis

Abstract:Objective To investigate the expression levels of microRNA-3163(miR-3163)and microfibrillar associated protein 2(MFAP2)in gastric cancer tissue and their relationship with clinical pathological parameters and prognosis.Methods From January 2016 to December 2020,A total of 92 gastric cancer patients were selected,and their general and clinical pathological data were collected.Cancer tissue and adjacent tissues were collected during surgery.Quantitative real-time PCR(qPCR)was applied to detect the expression of miR-3163.Immunohistochemical methods were applied to detect the expression of MFAP2.The relationship between the expression of miR-3163 and MFAP2 in gastric cancer tissue and clinical pathological parameters of patients were analyzed.The patients were followed up for 3 years,Kaplan-Meier was applied to analyze the relationship between miR-3163 and MFAP2 expression in gastric cancer tissue and prognosis.The influencing factors of prognosis were analyzed using Cox proportional hazards regression model.Results The expression level of miR-3163 in gastric cancer tissue(0.54±0.07)was lower than that in adjacent tissues(1.02±0.13),and the positive rate of MFAP2(57.61%)was higher than that in adjacent tissues(11.96%),and these differences were statistically significant(P<0.05).The expression of miR-3163 and MFAP2 in gastric cancer tissues were associated with TNM stage,lymph node metastasis,depth of invasion,and differentiation degree.In stage Ⅲ-Ⅳ gastric cancer tissues,as well as those with lymph node metastasis,deep invasion,and poor differentiation,the proportion of low miR-3163 expression and positive MFAP2 expression was significantly higher(P<0.05).Kaplan-Meier survival analysis revealed that the 3-year survival rate of patients with low miR-3163 expression was 58.33%,lower than 86.36%of patients with high miR-3163 expression(P=0.004).Similarly,the 3-year survival rate of patients with positive MFAP2 expression was 58.49%,lower than 89.74%of patients with negative MFAP2 expression(P=0.001).Multivariate Cox hazards regression analysis indicated that lymph node metastasis,deep infiltration,poor differentiation,low miR-3163 expression,and positive MFAP2 expression were independent risk factors affecting patient prognosis(all P<0.05).Conclusion In gastric cancer tissues,the expression of miR-3163 is downregulated while that of MFAP2 is upregulated.Both are closely associated with TNM stage,lymph node metastasis,depth of invasion,differentiation degree and poor prognosis,suggesting their potential as biomarkers for prognostic evaluation in gastric cancer.
