Expression of paraoxonase 2 in gastric adenocarcinoma tissues and its clinical significance

Abstract:Objective To investigate the expression of paraoxonase 2(PON2)in gastric adenocarcinoma tissues and its clinicopathological significance.Methods The mRNA level of PON2 in gastric cancer tissue was analyzed by bioinformatics.The expression difference of PON2 protein in gastric adenocarcinoma and normal gastric mucosa was analyzed by immunohistochemistry,and the relationship between PON2 protein and clinicopathological characteristics and prognosis of gastric adenocarcinoma was analyzed.Results Bioinformatics analysis showed that PON2 was significantly overexpressed in gastric cancer tissues(P<0.05).The immunohistochemical results showed that the high expression rate of PON2 protein in gastric adenocarcinoma tissue was 53.5%(46/86),higher than the 23.3%(20/86)in normal tissue adjacent to the cancer,and the difference was statistically significant(χ2=16.620,P<0.001).The differences in PON2 expression among different T stages(P=0.022),N stages(P<0.001),and TNM stages(P=0.018)of gastric adenocarcinoma were statistically significant.Gastric adenocarcinoma patients with high PON2 expression had a significantly shorter median overall survival of 37 months compared to 62 months of patients with low PON2 expression(P<0.001).Cox proportional hazards regression analysis demonstrated that high PON2 expression was an independent risk factor for patients'prognosis(HR=2.140,95%CI:1.108-4.131,P=0.023).Conclusion High expression of PON2 in gastric adenocarcinoma is significantly associated with tumor infiltration depth,lymph node metastasis,and clinical staging in gastric adenocarcinoma patients.PON2 may be a potential prognostic biomarker and therapeutic target for gastric adenocarcinoma.
